Design Notes
We based our design on the natural promoter Part:BBa_K822000, keeping the disposition of the operators and changing the strength of the promoter using Aderson promoters. This way we created:
- Part:BBa_K1847007, which contains a strong promoter Part:BBa_J23100
- Part:BBa_K1847008, which contains a medium promoter Part:BBa_J23118
- Part:BBa_K1847009, which contains a weak promoter Part:BBa_J23117
Then, we decided to change the arrangement of the operators to find out how this will affect the behavior of our system. We tried to put both operators next to each other following a promoter of different strength.
- Part:BBa_K1847002, which contains a strong promoter Part:BBa_J23100 followed by the two operator sites one next to the other.
- Part:BBa_K1847003, which contains a medium promoter Part:BBa_J23118 followed by the two operator sites one next to the other.
- Part:BBa_K1847004, which contains a weak promoter Part:BBa_J23117 followed by the two operator sites one next to the other.
